Effect of Acute Ethanol Intoxication on the protein, GSH and GSSG Contents of the Rat Kidney


Abstract
An effort was directed to elucidate the possible changes in the contents of protein, and glutathione(both reduced and oxidized forms) of the kidney in the acute alcohol intoxication.
The albino rats were used throughout the entire experiment, and 1.5 ml of 50% ethanol per 100 gm. BW was orally intubated, and the experiment was performed at 1, 3, 6, 12, and 24 hrs after the alcohol intubation.
The control was set in which the same amount of saline in place of alcohol was intubated, and otherwise the procedure was identical with the alcohol group.
The rat was sacrificed by puick blood shedding after cutting the both carotid arteries, and the kidneys were immediately excised and placed in the ice-cold saline solution until ready for use.
The protein content was measured by the method of Lowry et al, GSH by Ellman¢¥s method and GSSG by the electrolytic method of Dohan and Woodward. The results thus obtained are compared with the normal values and summarized as follows.
The kidney protein content of the rat in the normal group was 169.5¡¾5 98n-i"-/91-n, whereas in the alcohol group, the protein content of the kidney showed a tendency of increased values throughout the experiment.
The normal GSH value of the kidney was 4.78¡¾0.29pmol/gm, whereas in the alcohol group, significantly lower values were observed, i.e.; 3.87¡¾0. 17pmol/gm (p<0. 01) at 1 hr, 3.02¡¾0.25tpmol/gm (p<0.01) at 3 hr, 3.33¡¾0.31pmol/gm (p<0.01) at 6 hr, 3.50¡¾0.35jimOl/ gm (p<0.05) at 12 hr and 3.47¡¾0.21pmol/gm (p<0.01) at 24 hr.
The GSSG value of the normal kidney was 6.16¡¾0.50umol/gm, whereas in the alcohol group, a tendency of increased values from the normal were observed, but all the values were not statistically significant.
All the values in the control group did not show any significant change from the normal values.
